For Top Down Analysis experiments, you can add, delete, or edit target sequences using the Sequences Added to Experiment and Global Sequence Reference tables as necessary.

Add a new sequence to the Global Sequence Reference table

Procedure

  1. Select New Protein next to the Global Sequence Reference table.
  2. The Protein Sequence Editor opens.
  3. In the Protein Sequence Editor, import a FASTA file containing a new sequence of interest or manually enter the sequence and assign the Category as "Top Down".
  4. Save the sequence and close the Protein Sequence Editor.
  5. The sequence is added to the Global Sequence Reference table.

Edit an existing sequence in the Global Sequence Reference table

Procedure

  1. Select the sequence row in the Global Sequence Reference table.
  2. Select Edit next to the Global Sequence Reference table.
  3. The Protein Sequence Editor opens and displays the sequence information.
  4. In the Protein Sequence Editor, edit the sequence information as necessary.
  5. Save the sequence and close the Protein Sequence Editor.
  6. The sequence is updated in the Global Sequence Reference table.

Delete a sequence from the Global Sequence Reference table

Procedure

  1. Select the sequence row in the Global Sequence Reference table.
  2. Select Delete next to the Global Sequence Reference table.
  3. A confirmation dialog box appears.
  4. Select OK to delete the sequence.
  5. The deleted sequence is removed from the Global Sequence Table.

Add a sequence from the Global Sequence Reference table to the experiment

Procedure

  1. Select the sequence row in the Global Sequence Reference table.
  2. Select Add to Experiment next to the Global Sequence Reference table.
  3. The sequence is added to the Sequences Added to Experiment table.

Edit a sequence after adding it to the experiment

Procedure

  1. Select the sequence row in the Sequences Added to Experiment table.
  2. Select Remove next to the Sequences Added to Experiment table.
  3. Then, select the sequence row in the Global Sequence Reference table, select Edit, edit the sequence information as necessary using the Protein Sequence Editor, save the sequence, and re-add the sequence to the experiment.
  4. The edited sequence is re-added to the experiment.
  5. IMPORTANT

    If you edit any protein sequence added to the Sequences Added to Experiment table, you must return to the Component Detection subpage of the processing method editing wizard and reselect the protein sequence for each peak.

Remove a sequence from the experiment

Procedure

  1. Select the sequence row in the Sequences Added to Experiment table.
  2. Select Remove next to the Sequences Added to Experiment table.
  3. The sequence is removed from the Sequences Added to Experiment table.

Display the full sequence added to the experiment

Procedure

  1. Select the sequence row in the Sequences Added to Experiment table.
  2. Select Show Details next to the Sequences Added to Experiment table.
  3. The application displays the entire sequence in a dialog box.
