You can acquire sample data with the Tune application by setting the data acquisition parameters in the Tune window or by loading the parameter settings from an instrument method.
Procedure
- Open the Tune application.
- Do one of the following:
- Select the Ion Source tab and specify the ion source settings in the Ion Source pane. Then, select the Define Scan tab and specify the scan settings in the Define Scan pane.
- –or–
- Define the ion source and scan settings by selecting an instrument method in step 5.
- Place the MS in On mode.
- Select Continuous Acquisition to open the dropdown acquisition options pane.
- By default, the Continuous Acquisition check box is selected and the Instrument Method check box is clear. In addition, the Manual Stop option is selected for the Continuous Acquisition mode.
- Selecting the Instrument Method check box clears the Continuous Acquisition check box and changes the text on the Continuous Acquisition button to Instrument Method.
Data acquisition pane - Do one of the following:
- Keep the default setup for data acquisition.
- With the default data acquisition settings, you must select Stop
to stop data acquisition.
- Modify the setup for the Continuous Acquisition mode by selecting the Scans option or the Minutes option. Then in the box at the right, type the number of scans to acquire or the number of minutes to acquire scan data, respectively.
- When you select the Scans option or the Minutes option, data acquisition automatically stops after the MS acquires the specified number of scans or the specified time elapses, respectively.
- Select the Instrument Method check box. Browse to and select an instrument method. Then, select the Start Mode—Immediate, Contact Closure, or Divert Valve.
- When you select the Instrument Method mode, data acquisition automatically stops at the end of the method.
- (Optional) Select a different destination folder, specify a custom file name for the raw data file, and add more information to the raw data file as follows:
- To change the destination folder for the raw data file, select the Browse icon. Then, browse to the destination folder where you want to store the raw data files.
- The default folder location is drive:\Thermo\Data.
- In the File Name box, type a custom file name for the raw data file.
- The default file name is Rawdata with an appended time stamp.
- If the base file name already exists in the storage location that you specified, the Tune application adds a time-stamp suffix that consists of the year (YYYY), month (MM), day (DD), and time (HHMMSS).
- In the Sample Name box, type the name of the analyte (or other suitable label).
- In the Comment box, type a comment about the experiment.
- For example, describe the ionization mode, scan type, scan rate, sample amount, or method of sample introduction. The data system includes the comment in the header information for the raw data file.
- For the Continuous Acquisition mode, select Record
to start data acquisition.
- After the Tune parameters reach their specified settings, the data acquisition process begins and the following text appears below the Record icon—Recording in Progress.
- When you are ready, select Stop
to stop data acquisition.
