Prerequisites

1. Install the low-flow ESI spray insert. See Remove or Install the API Spray Insert.
2. Set up a 500 µL syringe with the FlexMix calibration solution. See Set up the Syringe.

Procedure

1. In the Tune application, place the MS in On mode .
2. Select the Profile icon and select a data type (profile or centroid).
3. Select the Positive (Negative) icon and select the positive ion polarity mode.
4. Select the Syringe Off icon to turn on the syringe pump.
5. The button name changes to Syringe On.
5. Select the drop-down arrow to open the syringe pump settings. Enter the following parameters and then select Apply.
6. - Flow Rate (µL/min): 3
7. - Volume (µL): 500

6. Verify that the inlet plumbing connections do not leak.
7. In the syringe pump settings, press and hold Prime to prime the syringe.
8. The default priming flow rate is 100 µL/min.
