You can manually acquire sample data with the Tune application by setting the data acquisition parameters in the Tune window or by loading the parameter settings from an instrument method.

Procedure

  1. Open the Tune application. For instructions, see Tune Application Window.
  2. Do one of the following:
  3. Select the Ion Source pane and specify the ion source settings. Then, select the Define Scan pane and specify the scan settings.
  4. Define the ion source and scan settings by selecting an instrument method in step 5.
  5. Place the MS in On mode.
  6. Select Continuous Acquisition to view the dropdown acquisition options.
  7. By default, the Continuous Acquisition checkbox is selected and the Instrument Method checkbox is clear. In addition, the Manual Stop option is selected for the Continuous Acquisition mode.
  8. Selecting the Instrument Method checkbox clears the Continuous Acquisition checkbox and changes the text on the Continuous Acquisition button to Instrument Method.

  10. Do one of the following:
  11. Keep the default settings for data acquisition.
  12. NOTE

    With the default data acquisition settings, you must select Stop to manually stop data acquisition.

  13. Setup the Continuous Acquisition mode as follows:
  14. - Select the Continuous Acquisition checkbox.
  15. - Select Scans and then enter the number of scans to acquire.
  16. - Select Minutes and then enter the number of minutes that you want data to be acquired for.
  17. NOTE

    The acquisition automatically stops after the MS acquires the specified number of scans or the specified minutes elapses.

  18. Setup an instrument method as follows:
  19. - Select the Instrument Method checkbox.
  20. - Browse to and select an instrument method.
  21. - Under Start Mode, select one of the following: Immediate, Contact Closure, or Divert Valve.
  22. NOTE

    When you select the Instrument Method mode, data acquisition automatically stops at the end of the method. If the instrument method contains both LC and MS parameters, the Tune window will only use the MS parameters because it does not control or inject a sample from the LC.

  23. (Optional) Select a different destination folder, enter a custom file name for the raw data file, and add more information to the raw data file as follows:
  24. To change the destination folder for the raw data file, select Browse . Then, navigate to the destination folder where you want to store the raw data files.
  25. The default folder location is C:\ProgramData\Thermo Scientific\Instruments\Data.
  26. In the File Name box, enter a custom file name for the raw data file.
  27. The default file name is Rawdata with an appended time stamp.
  28. NOTE

    If the base file name already exists in the storage location that you specified, the Tune application adds a time-stamp suffix that consists of the year (YYYY), month (MM), day (DD), and time (HHMMSS).

  29. In the Sample Name box, enter the name of the analyte (or other suitable label).
  30. In the Comment box, enter a comment about the experiment.
  31. TIP

    Describe the ionization mode, scan type, scan rate, sample amount, or method of sample introduction. The data system includes the comment in the header information for the raw data file.

  32. For the Continuous Acquisition mode, select Record to start data acquisition.
  33. After the Tune parameters reach their specified settings, the data acquisition process begins and the following text appears below the Record icon: Recording in progress.
  34. When you are ready, select Stop to stop data acquisition.
