You can view the results of a hydrogen deuterium exchange (HDX) experiment on the Hydrogen Deuterium Exchange page. The Hydrogen Deuterium Exchange page displays the HDX protection factor plots for each chain in the protein sequence in the experiment. This page is active only when you enable the HDX feature on the Hydrogen Deuterium Exchange page of the processing method editing wizard.

Procedure

  1. Open the results of your experiment from the Queue page or from the Load Results page.
  2. See Open results.
  3. The Process and Review page opens by default.
  4. In the navigation bar, select the Mapping tab.
  5. By default, the application opens the Coverage page under the Mapping tab.
  6. In addition to the Coverage page, the Mapping tab also contains the Modification Summary, Hydrogen Deuterium Exchange (for an HDX experiment), and Host Cell Protein Quantitation (if the HCP search feature is enabled) pages.
  7. On the Mapping page, select the Hydrogen Deuterium Exchange page subtab.
  8. The Hydrogen Deuterium Exchange page opens, displaying an HDX plot for each chain in the protein sequence used for the HDX experiment, with the title of each chain in red above the plot.

  10. For an experiment with only one raw data file, the plot shows only one HDX trace (one color and symbol).
  11. For an experiment with multiple raw data files, the plot shows a different color and symbol for each condition, with the color/symbol legend at the upper right of the plot.
  12. The numbers at the top of each plot represent the residue numbers in the sequence.
  13. The letters at the bottom of each plot represent the amino acids in the sequence.
  14. The left side of the plot displays the protection factor range for the entire plot. Each data point on each HDX trace shows the protection factor value for a particular amino acid and residue.
