Prerequisites

  1. You are viewing the Oligonucleotide Workbook and Calculator Editor.

Procedure

  1. In the Oligonucleotide Calculator and Editor, in the Oligonucleotide Workbook Parameters area, select the Manual Sequence subtab (if necessary).
  2. Type the name of the new sequence in the Chain Name box.
  3. TIP

Chain names can contain up to 250 alphanumeric characters.

  4. Type or paste the sequence chain into the sequence box, using either single-base notation (ATCGA) or triplet notation (Ad-pTd-pCd-pGd-pAd).
  5. Adding oligonucleotide sequences to the oligo workbook manually

  6. NOTE

A FASTA file might contain comment lines that begin with a greater-than sign (>), similar to the chain name line.

When you copy a chain sequence, do not include these additional comment lines. The application interprets comment lines as sequence building blocks.

  7. Select Apply.
  8. The Results table updates with the theoretical components for the new sequence added.
  9. Repeat this process to add additional sequences as needed.
