Use the following procedure to prepare 2 L of the regenerant (0.15 M TFA) from 99% TFA.

Procedure

  1. Add 1980 mL purified H2O (≥18 MW-cm) to a 2 L graduated cylinder.
  2. Stir the water with a magnetic stirrer, and add 23.2 mL (34.3 gm) of 99% TFA.
  3. NOTE

    A 500 mL bottle of 99% TFA will make ~ 43 L of 0.15 M TFA.

  4. Stir until solution is completely mixed, and transfer into a 2 L (P/N 39163), or 4 L (P/N 39164) reservoir for pneumatic delivery as the Dionex ERD 500 regenerant.

Because TFA is more volatile than H2O, any leakage of regenerant into the eluent stream will not result in concentration of leaked regenerant during evaporation. Since excess acidity may cause desialylation of oligosaccharides, the use of TFA regenerant is recommended for desalting of HPAEC/PAD separated sialylated carbohydrates.
