You can manually integrate component peaks in the selected ion chromatogram (SIC) for any single raw file data using the Manual Integrate button on the command bar on the Process and Review page.
Procedure
- Open the results of your experiment from the Queue page or from the Load Results page.
- The Process and Review page opens by default and displays the results of your oligonucleotide analysis experiment, including the chromatograms.
- In the Chromatogram pane, select the Enable Manual Integration (SIC only) checkbox.
- Select a component row in the Results table to populate the Chromatogram pane with the base peak (BPC) and selected ion (SIC) chromatograms for the component.
NOTE
You can manually integrate component peaks for a single raw file data only.
For an experiment with multiple raw data files, expand (+/− icon) a component row in the Results table and select the row for a single raw data file to perform manual integration.
- The selected component peak(s) are shaded in green on both the BPC and SIC.
- Using your cursor, select the peaks you want to manually integrate by defining the range of retention time (RT) start and stop times on the x-axis of the SIC for selected component.
- A red box appears around the selected RT range.
- Select the Manual Integrate button on the command bar to update the information on the Process and Review page.
- The selected RT range is shaded in blue on both the BPC and SIC.
- In the Results table, the Integration Type column is updated to "Manual" from the default value of "Automatic".
- The MS Area column (and the columns that depend on the MS Area) updates accordingly.
- The Trend Ratio and Trend MS Area plots ( which rely on the MS Area) update accordingly.
- On the Mapping page, the Coverage Map and Modification Summary update accordingly.
